With a vacuum manometer, you do need one line per carb.
Jumping around checking pairs does not work on a 4 cyl as you need to balance the vacuum between all carbs at once with a manometer.
( this is not the same as measuring specific vacuum values as you would do with a proper set of vacuum gauges)

on the 4 port manometer, do the lines need to be equal length to the "T" ? The curve of the outside lines would make them longer than the ones going into the T, yeah?
Would it be easier to ensure equal length if you put elbows on the outside so that the bottom is equal? for example:

ON ANOTHER NOTE,
for the electrolysis, can you put a whole tank in a plastic storage tub and do this same thing? would the rust travel out from inside the tank and to the sacrificial piece
 
Depending on the set up, it is possible to balance a 4 by doing 2 carbs at a time but it's time consuming and a PITA
CBX 6 cyl is done as two three cylinder motors then as a two cyl to get left and right banks matched
Actual line length isn't too important, just fit 1/16" restrictors in the fixed part of tube.
Old air jets work to slow down the movement of fluid, smaller than 60 makes things go too slow though
 
Another one - you need your own daytime show on the speed channel or something dude. This is awesome!

Ichiban, that is another great video, you make it look so easy but I don't know if I would want to attempt to do that myself. I don't have a measuring device and from your video it looks like the measurements are critical.
 
I'm selling those kits for $600 - for and additional $1000 you can upgrade to 250 µm tape. I have extremely reliable data that show this additional tape thickness adds over 500bhp to any motorcycle made before 1985.5.
