If i take of the stock 4-2s on my 1979 cb650 and replaced it with a 4-1 will i have to re-jet my carbs?

Maybe? Carbs are a guess and check game, unless you've done it thousands of time.

Start by setting your pilot jet adjustments, then setting your mains, then fiddling with the needles. Make sure everything is nicely synced too.

number one thing :
make sure the engine is in a PERFECT state of tune, that is all tuning procedures applied and running perfectly good,with the stock pipes,then swap out the pipes and tune the carbs with jetting changes if needed
