On a 1973 CB750, I'm using dual disk brake rotors on the front.
I have painted the hub, and both disks...
I'm thinking that I should sand ALL of the paint from the mating surfaces???

IMO, it's probably fine to leave on, but that really depends on how nice the bike is and what you want quality wise. Leaving it could cause chipping and mess up the painted surfaces.
I wouldn't worry about it.
Some bikes I have worked on had a heavy paper cushion between the rotors and the hub, presumably for damping vibration that would cause the brakes to squeal.
